Content

The theoretical part includes:

  • A thorough review of gene technology methods
  • Cellular control of macromolecular biosynthesis with emphasis on the experimental basis
  • Generation of genome- and cDNA libraries
  • Screening of libraries including expression cloning in Xenopus oocytes and micro organisms
  • Genome-, trancriptome- and proteome analysis
  • Production of transgenic animals
  • Heterologous expression in bacteria, yeast/fungi, insect cells, mammalian cells and whole animals


The practical part includes:

  • The most important methods to generate recombinant plasmids using restriction enzymes, Gateway® technology or homologous recombination
  • Site-directed mutagenesis
  • Real-time PCR for mRNA quantification and SNP analysis
  • Application of reporter genes to study gene regulation and subcellular protein localisation
  • Yeast 2-hybrid analysis
  • Affinity purification of fusion proteins
  • Pull-down experiments
  • Fluorescence microscopy
  • Internet based gene technology tools
  • Danish and EU law on gene technology
Learning Outcome

Knowledge:

The course provides the student with a detailed and comprehensive knowledge on theoretical and practical modern gene technology. The student gets aquainted with practical laboratory routines fulfilling the requirements stated by the Danish and EU law on gene technology.

Skills:

The student gains core competences within theoretical and practical modern gene technology. The degree of confidence with modern gene technology/molecular biology obtained during the course allows him/her to design gene technology/molecular biology experiments based upon known gene technological methods and internet based tools. The student obtains experience with critical interpretation and treatment of original molecular biology data, and gets acquainted with work routines in a gene technology laboratory in relation to Danish and EU law on gene technology and genetically modified organisms (GMOs).
Practical: The student gets aquainted with practical laboratory routines fulfilling the requirements stated by the Danish and EU law on gene technology.
Personal: The student learns to collaborate in groups and prepare a written presentation and discussion of their obtained experimental data.

Competencies:

The competencies obtained by the student include

  • to interpret data from the scientific literature and put them in a molecular biology/gene technology context
  • to design their own experiments
  • to work in a classified laboratory in accordance with legal regulations
  • to work in small teams
  • to give a detailed description of and evaluate the gene technology tools presented during the course
  • to apply the presented tools to design experiments with the potential to answer defined molecular biology questions
  • to describe and explain individual steps in a presented gene technology manual
